The U.S. Capitol has been surrounded by a security fencing perimeter since the January 6 riot of pro-Trump supporters who attempted to prevent the counting of electoral college votes for President Joe Biden.
William Evans, 18, has been named as the US Capitol police officer killed by the vehicle ramming.
The Congress is in recess and most lawmakers are away.
Friday's incident left at least one officer dead and another officer injured.
Police closed several blocks of Constitution Avenue, the main boulevard running along the north side of the US Capitol building.
They were called upon soon afterward to secure the Capitol during Biden's inauguration and faced another potential threat in early March linked to conspiracy theories falsely claiming Trump would retake the presidency.
Authorities have begun only in the past couple of weeks to remove the outer ring of high, razor-wire-topped fencing erected around the sprawling Capitol complex after the January 6 rampage.
The law enforcement officials told the Associated Press that the suspect was shot and was being taken to the hospital in critical condition.
"At that time, he started to lunge toward, run aggressively toward Capitol Police officers", Pittman said.
Both officers were hospitalised, and one of them "succumbed to his injuries", said Mr Pittman, who did not identify the slain officer or suspect.
In a tweet Friday, Senate Minority Leader Mitch McConnell said, "Praying for the United States Capitol Police officers who were attacked at the Capitol".
The security checkpoint is typically used by senators and staff on weekdays.
Video and images from the scene showed a dark coloured sedan crashed against a vehicle barrier and a police K-9 inspecting the vehicle.
Biden, who is spending the weekend at Camp David, also ordered the U.S. flag to be flown at half-staff at the White House and all federal buildings Friday until sunset on April 6 in honor of Evans.
